FREQUENTLY ASKED QUESTION?                                                                      http;//www.acinopeptide.com

What is peptides?
Peptides are short polymers of amino acids linked by peptide bonds. They have the same peptide bonds as those in proteins, but are commonly shorter in length. The shortest peptides are di-peptides, consisting of two amino acids joined by a single peptide bond. There can also be tripeptides, tetrapeptides, pentapeptides, etc. Peptides have an amino end and a carboxyl end, unless they are cyclic peptides.

Which analysis data is provided with the peptides?
Acinopeptide provides the Certificate of Analysis, HPLC and Mass data of peptides free of cost along with the peptides. We also provide other analysis data like NMR, MALDI-TOF, FTIR on request for an additional fee. GMP peptides include detailed records for each step of the process.

On what scale are the peptides synthesized?
We synthesize custom peptides in a range from mg up to several grams per peptide. Smaller aliquots are available upon request.

Does USV Peptides supply modified peptides?
Yes, we have a wide range of modifications available for peptides. For the complete list, please refer to our modifications table on this web site.

What purity levels are available?

We supply peptides having purity ranging from crude to >98% pur (By HPLC)

For peptide content, it is usually reach 85%

We control the Water content less than 10%

Also for the testing, we will provide full set analysis method and condition.

Peptide storage and use

How should I store my peptide?

For the most effective long term storage of peptides it is advised to store as a lyophilised powder at -20 C. Repeated freeze-thaw should be avoided, so if only small quantities are required at a time then aliquot before freezing. We can aliquot your peptide into smaller quantities for you on request, e.g. 10mg split into 1mg aliquots. Exposure to moisture will greatly decrease long-term stability of lyophilised peptides. To prepare the peptide for use, remove from cold storage and allow the peptide to equilibrate to room temperature before removing the lid of the container.

How should I solubilise my peptide?

First try to dissolve a small amount of the peptide in either water or buffer. If the peptide does not dissolve try using an organic solvent such as DMSO to solubilise the peptide, then dilute using water or buffer. Peptides containing cysteine are easily oxidised, so solutions should be prepared just prior to use. If your peptide requires special treatment you will be informed when the peptide is sent to you.

How do I make payment?

If you are new customers, we will ship your order once your payment is received.
If are you upgraded our star customers, we will give you different credit limit from USD1 to USD20000.00
We have five grades for star customer. If you need more information regarding our credit limit, please contact with info@acinopeptide.com

We accept following payment methods
+ Credit Cards: Major credit cards
+ West Union payment
+ Bank transfer, for our bank account, please contact with financial@acinopeptide.com

How long will I receive my order?
+
For stock peptide, we will arrange the shipment within one week after the order date.
+ For custom-synthesis peptide, it will take 3-4 weeks to finish the production.

How do I place an order? or How does acinopeptide process my order?
+ New customers should contact Acinopeptide first for quotes;
+ Pepmetric will contact and send you quotes within 48 hours for your review;
+ Customers arrange 30% advance payment and PepMetric confirms your order and approximate delivery date by email;
+ Peptide or peptide array is then custom made and delivered as soon as the rest 70% payment is received.

How do I cancel the order?
+ For stock peptide, you can cancel the order within the next one day after the order date. For example, if you order one peptide on 2011-01-01, you can cancel it on 2011-01-02, after that date, you can’t cancel it, and also you can’t get the advance payment.
+ For custom-synthesis peptide, you can cancel the order within next two days. For example, if you order one peptide on 2011-01-01, the latest cancellation date is 2011-01-03, after that date, you can’t cancel it, and also you can’t get the advance payment.

What is the recommended peptide length?
Coupling efficiency in FMOC solid phase spot synthesis can be as high as 98%; to maintain high integrity, fidelity and purity of peptides on membrane, the peptide length should be no longer than 16 amino acids though our have successful experience of synthesizing 20-mer peptides on arrays.

How much peptide is synthesized per spot?
Depending on array density, a typical spot of 5mm diameter will have >50nmol peptides (50~150ug in mass, depending on molecular weight of peptides) by theoretical calculation and assuming a coupling efficiency of >98% per cycle.

How does Acinopeptide QC the synthesis of SPOTs peptides in the custom SPOTs service?
Several internal controls (positive and negative controls) are synthesized on arrays to ensure the quality of peptide arrays.

Can the peptide array can be regenerated?
Due to its strong bonding nature of peptides with cellulose membrance, the membrane might be regenerated up to several times (not for protease and kinase arrays). However, regenerated membrane must be pre-tested first by your detection method in the absence of probe (such as target protein) before re-use to ensure membrane stripping is complete.

What are detection methods?
In general, many detection methods can be employed to detect peptide arrays, including fluoroscence, autoradiography, chemilluminescence, and chromogenesis as long as your antibody (or target protein) can be conjugated with corresponding fluorochrome, radioisotope, or HRP/AP. If you want to reuse membrane, chromogenetic substrates are not recommended.

What does the peptide array membranes look like?
The peptides are synthesized in a grid on cellulose membrane and four corners are marked by pencil, and the first peptide will be clearly marked for membrane orientation. Also, coming with membrane is a CD in which a grid diagram with detailed sequences for each spot as well as instructions will be included.
